Check out his Screencast from Rich Hoeg. It shows what you can load onto a USB thumb drive.

It covers running a portable Webserver (Apache) and database (MySQL) and loading portable applications including a Wiki.

There are tons of portable applications that can run from a USB drive.

A practical application for this would be someone who may not have high speed internet connection or field personnel with patchy access to the internet. They can have all their content/support material loaded on a USB thumb drive. A internal training Wiki for example or a library of PDF support documents with a PDF reader.
